It does exploit the one new tile. The 2:4:6:5 tile that the mouse is over. Don't I get those resource in my city once I build the district? Shouldn't it tell me I will be getting that income?

It's already in your city. :)

When you build a district, the total output indicates the bonuses you will gain through the new exploitations. For your situation, you already have this tile (as an exploitation), and if you build a district at this place, it won't add you any bonuses. That's why the total output is empty.
