I know we're still in early days in terms of the press that we'll be receiving about this game but what does it play like? Is it going to be a more direct descendant of Dungeon of The Endless with wave based tower defense or is it going to play more similarly to something twin stick like Alien Swarm?

From looking from the trail it seems it will go very much the same way as the first one. Turrets can be placed and you have to "carry" the stone around. But the main diffrent is that you have realtime action for eachroom in a top down shooter stile (helldiver, alien swarm) like fasion.

it does look like a similar gameplay to "Alien swarm : Reactive drop" while having the same mecanics Dungeon of the endless has

I think that Helldivers and Alien Swarm are fairly close, you might want to take a look at Livelock which might be even closer in terms of gameplay mechanics.
