Do you make use of additional antivirus software like Avast, BitDefender, Avira, etc.? My guess is, it blocks Endless Space 2 folder(s). Try to whitelist and restart game.
No, just the Windows Defender that's built-in to Windows 10.
What about admin rights for Endless Space 2.exe? Did you assign (right click on Endless Space 2.exe -> compatibility-> admin rights)?

What past latest update 1.3.14 ?
Fixed a compatibility issue with machines using Intel Iris 5100 & Pro 5200 that resulted in an indefinite hang on loading Fixed compatibility issue with the following integrated graphic chipsets: HD Graphics 5000, 5100, 5200, 5300, 5600
Also double check the game is running in administrator mode. This would be a error that would pop up if the game is trying to work with memory that has a higher allocation than its own.
There is a patch that wen't live a few minutes ago that fixed a few strange crashes. I think its now 1.39
The last thing I would try is let steam uninstall and re-install es2 (I know this takes time, or at a minimum use steams verify game integrity) to make sure it was installed properly. Everybody else covered everything already :(
